چکیده
Chapter 5 CREB Glycosylation Moderates Phosphorylation-Dependent CREB Activity in Cultured Pancreatic Cell Lines. Summary. In vitro studies of CREB glycosylation found that the transactivation potential of CREB is inhibited by glycosylation. Whether O-GlcNAc glycosylation is sufficiently dynamic to play a role in cellular activity has been addressed by several previous studies performed in pancreatic -cell lines [1-6]. The pancreatic cells were chosen as a model system because of the high levels of OGT and O-GlcNAc glycosylation present in b-cells [5, 7]. Our work using cultured pancreatic cells has shown that CREB glycosylation is indeed dynamic and capable of effecting a decrease in CREB-dependent transcription.

Dynamic O-GlcNAc Modification Regulates CREB-Mediated Gene Expression and Memory Formation
The transcription factor cyclic AMP-response element binding protein (CREB) is a key regulator of many neuronal processes, including brain development, circadian rhythm and long-term memory. Studies of CREB have focused on its phosphorylation, although the diversity of CREB functions in the brain suggests additional forms of regulation. Here we expand on a chemoenzymatic strategy for quantifyin...
متن کاملCREB Inhibits AP-2α Expression to Regulate the Malignant Phenotype of Melanoma
BACKGROUND The loss of AP-2alpha and increased activity of cAMP-responsive element binding (CREB) protein are two hallmarks of malignant progression of cutaneous melanoma. However, the molecular mechanism responsible for the loss of AP-2alpha during melanoma progression remains unknown. METHODOLOGY/PRINCIPAL FINDINGS Herein, we demonstrate that both inhibition of PKA-dependent CREB phosphoryl...

Physiological studies confirm improvement of memory by Olibanum, a resin from Boswellia species, while little is known about the molecular mechanism by which it affects memory performance. Two master transcription factors, CREB-1 and CREB-2, regulate downstream memory-related genes expression, leading to the long-term memory potentiation. This study addresses the effects of Beta-boswellic acid ...

متن کاملImpaired CREB-1 phosphorylation in antifolate-resistant cell lines with down-regulation of the reduced folate carrier gene.
The human reduced folate carrier (hRFC) is the dominant transporter for the uptake of antifolates used in cancer chemotherapy. We have shown recently that decreased cAMP-responsive element (CRE)-dependent transcription contributes to the loss of hRFC gene expression in multiple antifolate-resistant cell lines. This was associated with markedly decreased levels of phosphorylated cAMP response el...
